The next topic I covered with Vince was manufacturing. At Analyst Day, and prior to Analyst Day I heard a LOT of buzz about Arrowhead building a manufacturing facility in Madison, but there was no official news out there that I knew of. I told Vince that I'm hearing a lot of rumors about Arrowhead building or planning on building a manufacturing facility in Madison and if he had any news that he could share with me. Vince confirmed that Arrowhead is currently building a small-scale manufacturing facility in Madison. They leased out approximately 15,000 - 20,000 square feet from Roche right next door to Arrowhead's R&D facility. This facility is intended to be used for small clinical scale drug development (phase 1 - 3, and possibly for orphan drugs??). They are in the process of trying to get FDA validation of the facility. Arrowhead currently has no plans to develop a commercial scale manufacturing facility. As far as costs are concerned, Vince confirmed that Arrowhead is able to fund the development of this facility with their existing funds.

Conclusion: great to know that Arrowhead is investing in small-scale manufacturing without a capital raise. This shows Arrowhead's committment to growing in the clinic with uncommon speed.

Had a telephone interview with Vince Anzalone on Friday. I wanted to follow-up with him regarding a few questions I had after Analyst Day. Some of the answers are quite revealing, and I will break up the interview in a few posts, so enjoy.

I told Vince that I wasn't entirely convinced of the argument they provided regarding their new "dimer" molecule. What I didn't buy was that 6 mg/kg of the dimer drug was equivalent to two 3 mg/kg doses of the monomer, particularly since the dimer has twice as many triggers. So I therefore wanted to know if there were any advantages with regarding to being able to skip a step when testing the dimer in the clinic. Vince's confirmed that with the dimer, Arrowhead can begin testing a combo drug immediately, whereas two monomer drugs would need to be first tested individually before you can test the combo. This is clearly a big advantage, as Arrowhead is able to save at least 9-12 months time in the clinic, and it also saves on costs for having to run two additional phase 1 safety trials. Since this info was not featured in the presentation, I suggested to Vince that this significant piece of news should be included in future presentation about the dimer. Vince further elaborated that the dimer allows for simultaneous release of both triggers, and that this can be important when you're trying to knock down two genes at the same time, as the desired effectiveness may not be achieved if the timing is off. Vince also stated that the dimer molecule has a similar configuration as their ARO-HBV drug that has two triggers, except that the dimer knocks down two genes, instead of one at two different locations.
Conclusion: the new "dimer" molecule not only allows Arrowhead to target additional diseases with simultaneous targeting of two genes, but they're able to save about a year's time in the clinic.

ich kann es mir vorstellen das die Medikamente bei Wirksamkeit schnell durchgewunken werden.(Fast Track) ARO-AAT hat auch schon Fast Track.

Ich vergleiche die Charts hier auch mit yahoo - dort sind gestern gut 2 Mio Aktien gehandelt, hier an der Nasdaq nur 510 000 - diese Unterschiede sieht man im Jahresverlauf häufig - was stimmt denn jetzt.

Es gibt 95 Mio Aktien. 69 % sind in institutionellen Händen, über 8 % bei Insidern.
Heißt: Es gibt nur ca 20 Mio Aktien im Freefloat. Das hieße ja bei dem Volumen, dass nach ca 15 Handelstagen alle Aktien einen neuen Besitzer haben.

Ich versuche nur mein Staunen über den Ausbruch nach oben in Verstehen zu wandeln. Anscheinend gibt es hier gerade eine grundsätzliche Neubewertung der Aktien.

die yahoo Zahlen werden wohl stimmen. Die Medikamente sind bei Zulassung so viel Wert das es den Kurs über 1000 Dollar heben wird? Die Plattform bietet riesiges Wachstumspotential, das wohl auch bald mit Partnern gehoben wird?
Hier ensteht ein Pharmariese, kann man auch an den Neueinstellungen sehen. Deswegen hebt die Aktie jetzt ab.
Denke ich mir mal so. :-)))

ja, ist sicherlich eine Absicherung von einigen Institutionellen- teils wohl auch einfach eine Wette - wenn es plötzlich doch eine starke Nebenwirkung gibt.
Teils wird ja auch unfair gekämpft und Zweifel gestreut - wir werden sehen - ist nur in meinen Gedanken, damit man psychisch vorbereitet ist. :)

Eine beschleunigte Zulassung kann schon während der Phase 2 erteilt werden. Theoretisch also schon in gut einem Jahr, wenn konkrete Ergebnisse vorliegen und nicht nur Zwischenergebnisse.

 

hier wird es keine starken Nebenwirkungen geben. Wir haben ein besseres Sicherheitsprofil als Alnylam und Medicincompany.

Montag kommen Daten von den beiden Medikamenten ARO-APOC3 und ANG3, die PCSK9 Killer so wie der Cheffe immer sagt :-)  

und deren Wertschöpfungsmöglichkeiten habe ich mich noch gar nicht gekümmert, obwohl - ich muss ja sagen - "dein" Thread hier durch dich bestens informiert ist.  

in Deutschland hat heute einen Medikamenten oder Placebomix gegen chronisch Hepatitis bekommen - bisher keine Nebenwirkungen.

Eine Korrektur in der Aussage des Facharztes: Ein Ethikrat aus Fachärzten beurteilt jetzt ständig, ob Nebenwirkungen stark sind und die Studie unterbrochen oder beendet wird - oder auch ob so wenig Nebenwirkungen bei hoher Wirksamkeit festzustellen sind, dass es unethisch wäre, kranken Patienten einen Placebo zu geben. Dann würde die Studie umgestellt werden und alle würden einen Medikamentenmix bekommen - keiner mehr Placebo. Das hält er bei den bisherigen Ergebnissen hier für möglich. - Dies hat aber nichts mit einer beschleunigten Zulassung zu tun. Diese muss nicht unbedingt hier eintreffen, da die chronische Hep ja sehr schleichend ist und der Virus mit bisherigen Medikamenten unterdrückt werden kann.

zu APOC3 und ANG3, dass sie hochwirksam sind und wenig bis gar keine Nebenwirkungen.

Das Potential bei dem Thema Stoffwechsel, Fette, Cholesterin sollte sehr hoch sein, aber ist eine subjektive Meinung und ich finde noch keine professionellen Vorausschauen.  

mit Cholesterinsenkern(Statine) werden jedes Jahr über 15 Milliarden Umsatz gemacht?

2014 ist das Medikament mit den drittstärksten Umsatz weltweit mit über 8 Mrd. ein Chol-Senker

und im nächsten Jahr die Phase 3 dafür.

warum das Hep B Medikament eine Art Burggraben hat und der Favorit der Anleger ist.
Sie erwarten bald nähere Infos von dem letzten  Leber-Kongress.
